The basic helix-loop-helix (bHLH) transcription factor is one of the most important gene families in plants, playing a key role in diverse metabolic, physiological, and developmental processes. bHLH family is composed of approximately 60 amino acids with two functional domains, namely a basic conserved motif and a HLH motif. The basic motif consists of approximately 17 amino acids and possesses the typical six basic residues at the N-terminal, which functions to bind to the E-box (5'-CANNTG-3') or the variant G-box (5'-CACGTG-3') element. The HLH motif is composed of two amphipathic α helices separated by a loop region with variable size, and the loop region could participate in the formation of homodimeric or heterodimeric complexes, which contribute to the specific binding of bHLH transcription factors on their target genes.
